Vesicular Transport: EnSNAREd by GAPs
نویسندگان
چکیده
How are SNARE proteins included into transport vesicles? One way is through the interaction with ArfGAP proteins. A recent study reports that the ArfGAP Hrb can wrap around the SNARE VAMP7, causing its endocytosis by clathrin-coated vesicles.

Retrograde transport from the yeast Golgi is mediated by two ARF GAP proteins with overlapping function.
ARF proteins, which mediate vesicular transport, have little or no intrinsic GTPase activity. They rely on the actions of GTPase-activating proteins (GAPs) for their function. The in vitro GTPase activity of the Saccharomyces cerevisiae ARF proteins Arf1 and Arf2 is stimulated by the yeast Gcs1 protein, and in vivo genetic interactions between arf and gcs1 mutations implicate Gcs1 in vesicular ...
متن کاملIdentification of the catalytic domains and their functionally critical arginine residues of two yeast GTPase-activating proteins specific for Ypt/Rab transport GTPases.
Ypt/Rab proteins constitute the largest subfamily of the Ras superfamily of monomeric GTPases and are regulators of vesicular protein transport. Their slow intrinsic GTPase activity (10(-4)-10(-3) min(-1) at 30 degrees C) has to be accelerated to switch the active to the inactive conformation. We have identified the catalytic domain within the C-terminal halves of two yeast GTPase-activating pr...
متن کاملTBC1D20 is a Rab1 GTPase-activating protein that mediates hepatitis C virus replication.
Like other viruses, productive hepatitis C virus (HCV) infection depends on certain critical host factors. We have recently shown that an interaction between HCV nonstructural protein NS5A and a host protein, TBC1D20, is necessary for efficient HCV replication. TBC1D20 contains a TBC (Tre-2, Bub2, and Cdc16) domain present in most known Rab GTPase-activating proteins (GAPs). The latter are mast...
متن کاملArfs at a glance.
The Arf small G proteins regulate protein and lipid trafficking in eukaryotic cells through a regulated cycle of GTP binding and hydrolysis. In their GTP-bound form, Arf proteins recruit a specific set of protein effectors to the membrane surface. These effectors function in vesicle formation and tethering, non-vesicular lipid transport and cytoskeletal regulation. Beyond fundamental membrane t...
متن کاملAdvantages and limitations of cell-based assays for GTPase activation and regulation
Small GTPases of the Ras superfamily are important regulators of many cellular functions, including signal transduction, cytoskeleton assembly, metabolic regulation, organelle biogenesis and intracellular transport. Most GTPases act as binary switches, being "on" in the active, GTP-bound state and "off" in the inactive, GDP-bound state, and cycle between the two states with the aid of accessory...
